I want each SKU to have just one barcode associated with it. How can I accomplish this?

There is an option in the System Config > Account Settings menu, Allow multiple barcodes per SKU. By default, this is checked. However, you can uncheck this option so that each SKU can only have one unique barcode.

If I uncheck the Allow multiple barcodes per SKU option, what happens if I receive an item to a SKU that currently has multiple barcodes?

When the receiver scans a barcode different than what is already stored for that SKU, they’re prompted with a message indicating such and will need to either edit the barcodes on the SKU and record the one they have in front of them, or bypass recording their barcode against that SKU. It will not allow them to scan a different barcode than one already loaded.
